Overview

RoosterHD™-EV is a highly productive, chemically defined scalable media system designed to maximize upstream exosome yields in bioreactors and simplify downstream purification. By using a single fed-batch media system from cell seeding through harvest, RoosterHD-EV extends traditional exosome collection windows and bypasses particle loss during media exchanges and washing steps.

Using RoosterHD-EV results in a 10 to 20-fold increase in exosome yields which leads to a significant reduction in manufacturing costs. The chemically defined formulation is designed to simplify downstream processing and reduce regulatory complexity.

2D flask and 3D bioreactor process recommendations support easy integration into manufacturing workflows and RoosterBio’s downstream process expertise and EV Analytical Services are available to further support program manufacturing, testing, and development needs.

Microcarriers not included:  Per our protocols and process recommendations for bioreactor cell culture, RoosterBio suggests using either Corning® Low Concentration Synthemax® II Microcarriers (3781) or Sartorius SoloHill Collagen-coated APC Microcarriers (C/CIR-221).

RoosterHD-EV enables 6-Day exosome production during cell growth.
RoosterHD-EV enable 12-Day exosome production in 3D bioreactors to achieve a 10 to 20-fold increase in yields.
hMSC exosomes produced in RoosterHD-EV exhibit hallmark MSC EV tetrospanins as measured by single particle nanoflow cytometry and capillary western blot.
hMSC exosomes produced in RoosterHD-EV exhibit both bioactivity measured by CD73 activity assay & immunomodulatory function measured by macrophage polarization assay.
