Overview

RoosterBio’s 3D Bioreactor Media Kit is an all-in-one system for 3D bioreactor expansion of MSCs, iPSC-MSCs, immortalized MSCs, and fibroblasts. Engineered for fed-batch expansion to remove costly media exchanges, enhance overall productivity, and reduce in-process contamination risks;  this product has been vetted in a variety of commercially available bioreactor technologies to simplify integration therapeutic development programs.

Comprised of RoosterNourishTM-MSC-XF expansion media and RoosterReplenishTM-MSC-XF concentrated bioreactor feed, this system reduces media costs by up to 70%.  For seamless transition to exosome collection, pair with one of our EV Collection Products.

GMP CliniControl products are available for clinical manufacturing. Contact your RoosterBio representative for information on cGMP Bioreactor Media options for large scale (3L to >50L) MSC or EV Production.

Microcarriers not included:  Per our protocols and process recommendations for bioreactor cell culture, RoosterBio suggests using either Corning® Low Concentration Synthemax® II Microcarriers (3781) or Sartorius SoloHill Collagen-coated APC Microcarriers (C/CIR-221).

Fig 1: A comparison of XF hMSC cell densities in bioreactor culture using batch and fed-batch processes. RoosterVial™-hBM-XF (Xeno-free human bone marrow-derived mesenchymal stem cells – RoosterVial™-hBM-10M-XF – were cultured in suspension on microcarriers in RoosterNourish™-MSC-XF and fed with RoosterReplenish™-MSC-XF on Day 3, and compared to batch culture.
Fig 2: Fed-batch culture using RoosterReplenish™-MSC-XF is the most efficient and productive culture feeding regimen, outperforming either 1/2 media exchange or batch culture in media productivity by over 70%, and achieving up to 350 million (M) cells/L. This productivity is >17 times the media productivity of traditional hMSC flask-based cultures of ~20M cells/L, and is the central driving force for cost reduction in bioreactor-based manufacturing. Note: Total cell yield is donor-dependent.
